Did you know that (ER) stress in the skin arises from dysregulation of ER calcium store and impact of reactive oxygen species caused by external disturbances, such as epidermal barrier perturbation, UV irradiation, heat, and other types of oxidative stress.

Dysregulation of ER

Disruption of ER calcium homeostasis leads to a depletion of calcium stores within the ER, resulting in the release of calcium that compromises the function of calcium-dependent chaperones and consequently causes protein misfolding.

Key role of cells

In response to ER stress, cells activate the Unfolded Protein Response (UPR), a mechanism aimed at alleviating stress and restoring ER function to support cell survival and adaptation. However, if ER stress persists and the UPR fails to resolve it, apoptosis, a programmed cell death process, may be triggered instead.

Preserving cells from excessive ER stress

Excessive ER stress can lead to an inability to restore normal ER function, which is associated with skin conditions such as Rosacea. Cosmetics could address this issue preserving skin cells from ER stress by minimizing calcium depletion and promoting the pro-survival UPR axis.
